Basically a hopper takes 0.4s to transfer one item. In a full cycle each item has to go there and back, so 0.8s per item. That means a full 5 stacks will give you 320x0.8s = 256s. That's not quite accurate as there is an additional delay whenever the pistons move. There's a setting in the calculator you linked to include that as well.
