Thanks to the DLC, we now know Ranni absolutely did not have to kill Godwyn and unleash the plague of deathblight upon the world to shed her empyrean flesh and her great rune. All she had to do was go to the land of shadow and do it herself like Miquella did.
Rennala is Ranni's adoptive mother, and she really was born to Radagon/Marika, and the twin thing is what caused the birth defects. Many different conditions exist that could qualify you as empyrean, and being born to a single god and nobody else is just one of them.
Ranni was simply chosen as an Empyrean like Marika and the Gloam Eyed Queen before her. We even know Marika was not always a Goddess/Empyrean, we were told this in the DLC description, where it explicitly says she became a Goddess in the Land of the Shadows.
