The county was organized in 1821 and named for U.S. Representative John Scott, the first federal representative from Missouri. Scott County comprises the Sikeston, MO Micropolitan Statistical Area, which is also included in the Cape Girardeau -Sikeston, MO- IL Combined Statistical Area.
The first American settlement in the area now known as Scott County was Clark's Ferry or Clark's Landing (now Buffalo) in 1833. Other early towns included Davenport (now the county seat and largest city) and the town of Rockingham (which ceased to exist in 1847).
The area of Scott County and much of southern Minnesota became available for European American re-settlement by two treaties signed at Mendota and Traverse des Sioux in 1851 and 1853, respectively. Savage, 1943. Land speculators and settlers entered the area in droves staring in the mid-1850s.

Settlers started entering the area in the mid-1850s. The Minnesota River and the ox cart trails were the primary transportation routes.
