- Mario. Of course, the film wouldn’t exist without the main star, Mario, voiced by Chris Pratt. The plumber-turned-hero is on a great new adventure, seemingly happening in the Mushroom Kingdom after falling through a pipe.
- Bowser. While Mario is the protagonist, his nemesis was the star of the first trailer. Bowser, the king of the Koopas, is being voiced by the great Jack Black, and the common consensus on Twitter is that this is perfect casting.
- Princess Peach. There was one glaring and strange omission from the trailer and that was the absence of Princess Peach. The Queen’s Gambit’s Anya Taylor-Joy is voicing the effervescent damsel-in-distress.
- Luigi. Fans were delighted to see Luigi pop up even briefly in the teaser, and naturally, he’s running away from his enemies. This is another case for perfect casting as It’s Always Sunny in Philadelphia’s Charlie Day is voicing Mario’s scaredy-cat brother, Luigi.

- Chris Pratt as Mario. Mario is a struggling Italian-American plumber based in Brooklyn, New York. When Luigi is captured by Bowser’s army, he sets out on a mission in Mushroom Kingdom to save his younger brother.
- Anya Taylor-Joy as Princess Peach. As the ruler of the Mushroom Kingdom, Princess Peach acts as Mario’s guide, mentor and love interest. She has lived in Mushroom Kingdom since she was an infant and was raised by the Toad population within the community.
- Charlie Day as Luigi. Luigi is Mario’s timid younger brother and business partner. After he is captured by Bowser and his army, Mario sets out on a mission to save Luigi.
- Jack Black as Bowser. Bowser is King of the Koopas, the ruler of the Dark Lands. In the movie, he steals the powerful Super Star in an attempt to conquer the Mushroom Kingdom.
