As the global obesity epidemic continues to escalate, the need for effective and safe weight loss interventions becomes increasingly pressing. The concept of using ETC uncouplers as a treatment for obesity remains intriguing, as it offers a unique approach to increasing energy expenditure and promoting weight loss.
Safe and effective pharmacological interventions are therefore needed. Liver-targeted mitochondrial uncoupling agents represent a unique therapeutic strategy to increase hepatic lipid utilization and reduce hepatic steatosis and its ensuing metabolic complications.
Jun 8, 2020 · Mitochondrial uncouplers transport protons into the matrix by bypassing ATP synthase, which throws off the PMF. To reestablish the gradient, protons must be exported out of the mitochondrial ...
Mitochondrial uncouplers such as 2,4-dinitrophenol (DNP) promote both weight loss and heat generation.1 HU6 is a prodrug, metabolised in the liver to produce DNP. Mazen Noureddin and colleagues2 reported a phase 2a clinical trial in people with non-alcoholic fatty liver disease (NAFLD) and high BMI, showing that HU6 decreased mean liver fat content by 26·8–35·6% and bodyweight by 0·52–2 ...
Classical uncouplers have five properties: [1] the complete release of respiratory control; the substitution of all coupled processes (ATP synthesis, transhydrogenation, reverse electron flow, active transport of cations, etc.) by a cyclic proton transport mediated by the uncoupler

Background. Mitochondrial uncouplers are well-known for their ability to treat a myriad of metabolic diseases, including obesity and fatty liver diseases. However, for many years now, mitochondrial uncouplers have also been evaluated in diverse models of cancer in vitro and in vivo. Furthermore, some mitochondrial uncouplers are now in clinical ...
