Abstract. Complex chromosome rearrangements, known as chromoanagenesis, are widespread in cancer. The most frequent type based on large-scale DNA sequencing of human tumors is chromothripsis, a massive, localized and clustered rearrangement of one (or a few) chromosomes seemingly acquired in a single event.
Complex Chromosome Rearrangement. Complex chromosome rearrangements have three or more breakpoints and may lead to a balanced or an unbalanced copy-number state [60,61]. Recent NGS breakpoint studies have paved the way to understanding the mutational mechanisms and defining the genomic structure of these rearrangements.
(a) Complex rearrangements of the short arm of human chromosome 17 revealed by FISH analysis (adapted, with permission, from Ref. ). (b) Complex rearrangement at 1q revealed by BAC aCGH. A BAC array is shown on the left with the chromosome numbers depicted above.

Complex genomic rearrangements have been reported in lymphoma. 40-45 Complex chromosomal translocations leading to gene fusions are also found in solid cancers, 19 including whole arm translocations and isochromosomes in head and neck squamous cell carcinoma, 46 complex genomic rearrangements including inversions in pancreatic cancer, 47 and rearrangements including tandem duplications in ...
